Immunoscreening of Schistosoma japonicum Adult Worm cDNA Library by Serum from Swine Vaccinated with Ultraviolet Attenuated Cercariae

Abstract

Objective To explore the antigenic molecules of ultraviolet radiated attenuated cercariae of Schistosoma japonicum,and get some future candidate antigens for developing vaccine and controlling measure against schistosomiasis.Methods Adult worm cDNA library of Schistosoma japonicum was screened by serum of swine vaccinated with ultraviolet attenuated cercariae,and the best cDNA library which was used for screening was detected titer.Results The best titer of cDNA library for screening was 1.89×109ρfu/mL,according to three rounds of screening,20,10 and 7 positive clones were obtained respectively at attenuated cercariae group,and at control cercariae group,15,9 and 3 positive clones were screened,but at normal serum group,no positive clones were screened.Conclusions The positive clones obtained maybe were the genes encoding the antigens which could provide protective immunity against schistosomiasis and elicit the directions for antigenic genes encoding antigen against Schistosoma japonicum.
