Abstract

[Objective]The research aimed to confirm the best extraction,alcohol precipitation and purification conditions of polysaccharide in Chrysanthemum morifolium Ramat.[Method]With the yield of polysaccharides as an index,the extraction,alcohol precipitation and purification processes of polysaccharide in C.morifolium were optimized.[Result]The influence sequence of the factors of water extraction of poly-saccharide from C.morifolium was as follows:extraction timessolid-liquid ratioextraction temperatureextraction time.The best exraction process was as follows:solid-liquid ratio of 20∶1(V∶M,g/ml) ,extraction time of 4 h,extracting at 95 ℃ for 3 times.The influencing sequence of alcohol precipitation process was alcohol precipitation timeethanol volume fractionconcentrated degree of medicine liquid.The alcohol precipitation process was as follows:ethanol volume fraction of 80 % ,concentrated degree of medicine liquid of 1∶ 3,alcohol precipitation time of 9 h.The influencing factors sequence of purification technology was as follows:chloroform/n-butanol purification timesamples/chloroform+n-butanol.The purification process was as follows:chloroform/n-butanol of 5∶1,the volume ratio of samples to chloroform and n-butanol of 2∶1,purification time of 10 min.[Conclusion]The process was suitable for the extraction of polysaccharide from C.morifolium.
