Effect of fluvastatin on renal transforming growth factor 1 in diabetic rats

Abstract

Objective To investigate the effect of fluvastatin on renal transforming growth factor 1(TGF1) in diabetes rats. Methods The following groups of rats were studied: normal control ratsstreptozotocin diabetic rats and diabetic rats treated with fluvastatin(2mgkg-1d-1).Plasma glucose, serum cholesterol, triglyceride and creatinine, urinary albumin as well as profile of kidney hypertrophy were observed after 2,8 weeks of treatment, while TGF1 mRNA expression of kidney contex was assessed by Northern blot analysis.TGF1,fibronectin and collagen protein expression were measured by immunohistological staining. Results Serum cholesterol,urinary albumin excretion(P001) and kidney hypertrophy index(P005)in diabetic rats of fluvastatin treated group were significantly lower than diabetic untreated rats.Compared to control group there was a significant increase in mRNA and protein expression of TGF1 in association with increase of protien expression of fibronectin and collagen in diabetes rats(P001).The mRNA expression of TGF1(P001) as well as the protein expression of TGF1fibronectin and collagen (P005)in fluvastatin group were much lower than diabetic untreated group. Conclusion Fluvastatin has some renal protective effect on diabetes rats, partly through downregulating TGF1 expression and reducing deposition of glomerular ECM.
