Abstract
Objective:To investigate the expression and significance of oncogene Pokemon in nasopharyngeal carcinoma(NPC),and reveal the association between NPC pathogenesy and clinical parameters.Methods:The expression of Pokemon protein was detected by using immunohistochemical techniques(S-P) in 86 cases of NPC,and 30 controls of nasopharyngitis tissues.Mean while,we explored the expression of Pokemon mRNA in 56 cases of NPC patients and 30 controls of pharynx nasalis chronic inflammation by using reverse transcriptase polymerase chain reaction(RT-PCR).Results:S-P method proved that Pokemon was highly expressed in 86 cases of NPC,and it located in the nuclear of NPC cells.The rate of strong positive expression in the NPC(67/86,77.9%) was significantly higher than in controls(7/30,23.3%,P0.05);The expression of Pokemon was significantly higher in lymph node metastasis group than no-metastasis group.In different clinical stage,the expression of Pokemon was different(P0.05).Pokemon mRNA was strongly amplificated in the NPC cases(51/56,91.1%),but not in the 30 cases of nasopharyngitis tissues(0/30,0%,P0.05).The rate of positive expression in early stage(Ⅰ+Ⅱ,35/39,89.7%) was lower than in the advanced stage(Ⅲ+Ⅳ,16/17,94.1%,P0.05).The relative expression levels of Pokemon mRNA were very different between lymph node metastasis group and no-metastasis group(P0.05).Conclusion:There was less expression of Pokemon in nasopharyngitis tissues than in nasopharyngeal carcinoma.Pokemon expression was correlated with the tumor stage and lymph node metastasis.Pokemon plays a vital role in the progression of NPC.Pokemon is likely to a molecule parameter in the diagnosis and clinic evaluation,and may be a potential target in anticancer of targeted therapy.
