Abstract

Objective To understand the expression of Fragile Histidine Triad Gene (FHIT) protein and Proliferating Cell Nuclear Antigen (PCNA) in normal cervical epithelia,cervical intraepithelial neoplasia (CIN) and cervical cancer,and to explore their clinical significance in the early diagnosis and the development of cervical cancer.Methods The expression of FHIT protein and PCNA were detected by immunohistochemical method in 20 cases of normal cervical epithelia,20 cases of CINⅠ,25 cases of CINⅡ/Ⅲ as well as 60 cases of cervical squamous cell carcinoma.Results Both the negative rate of FHIT expression and the positive rate of PCNA expression increased from normal cervical epithelia,CINⅠ,CINⅡ/Ⅲ to cervical cancer with statistical significance.Additionally,FHIT expression deletion was associated with pathological grade,lymph node metastasis.However,there were no relationship between PCNA expression and age,tumor size,pathological grade,clinical stage as well as lymph node metastasis.The expression of FHIT protein in cervical cancer showed a negative correlation with PCNA expression (r=0.193).Conclusion The expression level of FHIT is associated with the grade of CIN,so it would be used to screen or predict CIN.Additionally,FHIT and PCNA are valuable for screening and early diagnosis of cervical precancerous changes and cervical cancer.
