Abstract
Objective To explore methods whether the Insulin Resistance model can be established through feeding high-sugar and high-fat diet in Wistar rats. Methods 24 Wistar rats were randomly divided into control group (feed with common diets) and model group (feed with high-fat and high-sugar diets). After 12 weeks, rat's blood was collected, and fasting plasma glucose, fasting plasma lipids and insulin levels were detected by biochemistry or ELISA technique to evaluate the effects of different diets on insulin sensitivity index (ISI) and insulin resistance index (HOMO-IR). Results The serum low density lipoprotein (LDL-C), total cholesterol (TC), triglyceride (TG2), fasting plasma glucose (FPG) of the model group was significantly increased, comparing with the control group(P 0.05). Fasting insulin levels (FINs), insulin sensitivity index (ISI), insulin resistance index (HOMO-IR) were also significant differences between two groups, such as ISI decreased obviously (P0.05), while the HOMO-IR increased significantly (P 0.05), comparing with the control group. Conclusion We could obtain a stable high glucose and high fat rat model through feeding Wistar rat with high-sugar and high-fat diet, and may be able to initially established the insulin resistance animal model with hypoinsulinism, rather than a single animal model of insulin resistance. This animal model of insulin resistance occurred in line with the law of humanity, it is an ideal animal model.
