Abstract
Objective To study the role of Fhit、Caspase-8 in the development of carcinoma of bladder,as well as its clinical significance.Methods The expressions of Fhit and Caspase-8 were measured by immunohistochemistry in the normal bladder samples、cystitis glandularis、paraneoplastic samples and carcinoma of bladder,and their average optical was detected by image analysis system.Results The average optical of Fhit and Caspase-8 in normal bladder samples、cystitis glandularis、paraneoplastic samples、transitional cell carcinoma of bladder and adenocarcinoma of bladder were 0.270±0.042、0.248±0.044;0.142±0.040、0.150±0.051;0.240±0.061、0.234±0.053;0.097±0.053、0.108±0.053;0.089±0.026、0.105±0.062 respectively.The expression of Fhit and Caspase-8 in carcinoma of bladder was significantly lower than that in normal bladder samples(P0.01).The intensity of Fhit and Caspase-8 expression was decreased with the increasing of grades of tumor(P0.01),and obviously correlated with the grades respectively(r=-0.700、r=-0.674,P0.01),but incorrelated with stages of tumor(r=0.456,P=0.025;r=0.337,P=0.107).A positive correlation was found between Fhit protein loss and Caspase-8 reduced expression(r=0.726,P0.01).Conclusion The down regulation of Caspase-8 in bladder neoplasm in connection with Fhit protein loss reveals the close relationship between Fhit 、 Caspase-8 and the occurrence and progression of the cancer.
