Abstract
[Objective] The purpose was to discuss the culture condition and partial characteristics for producing chitinase with marine fungi HZ-01 strain.[Method] A strain of fungi HZ-01 with high yield of chitinase was screened out from marine environment,with which as material,the experiment on the influence of different carbon(C) source,nitrogen(N) source and temperature on producing chitinase with the strain through fermentation was done to study the culture condition of chitinase.After the crude enzyme solution was treated with heat preservation for 60 min under different temperatures,the samples were fetched to determine chitinase activities and analyze the heat and acidic-basic stabilities of chitinase.[Result] The optimum C source for producing chitinase with marine fungi HZ-01 strain was chitin and peptone was the optimum N source.When the temperature was 30 ℃,with 1.0% chitin as C source and 0.1% peptone as N source,the produced chitinase had highest activity and the enzyme activity decreased along with the increment of temperature.The acidic-basic stability of the enzyme was worse,which only had higher activity in pH value of 6~8.[Conclusion] The research laid primary foundation for scale production of chitinase.
