Behavioral Effects of Different Dose of MK-801 in Reserpine Induced Behavioral Depressive Rats

Abstract

Objective:To investigate the behavioral effects of different dose of N-methyl-D-aspartate receptor antagonist MK-801 in reserpine-induced behavioral depressive rats.Methods:Fourty male SD rats were randomly divided into control group,reserpine(4 mg/kg)group,MK-801(0.1、0.3、0.5 mg/kg) + reserpine(4 mg/kg)group.The depressive-like behaviors were checked by open field test and forced swim test.Results:One hour after intraperitoneal injection of reserpine(4 mg/kg),the open field test showed that the total distance and the distance travelled in central zone of reserpine group were significantly decreased compared to the control group(P0.01),the time travelled in central zone was decreased(P0.05).The total distance of MK-801+reserpine group was significantly increased compared to the reserpine group(P0.01).The distance and time travelled in central zone of 0.3、0.5 mg/kg MK-801 + reserpine group were increased compared to the reserpine group(P0.05).The distance travelled in central zone of 0.3、0.5 mg/kg MK-801+reserpine group was increased compared to the 0.1 mg/kg MK-801 + reserpine group(P0.05).The ataxia adverse effect was observed in 0.5 mg/kg MK-801 + reserpine group in open field test.The forced swim test showed that the immobility time of reserpine group was significantly increased compared to the control group(P0.01),the immobility time of MK-801+ reserpine group was significantly decreased compared to the reserpine group(P0.01).Conclusion:MK-801 can antagonize the behavioral depression caused by intraperitoneal injection of reserpine(4 mg/kg),and 0.3 mg/kg MK-801 is a suitable dose in this test.
