Study on extraction and analysis of polysaccharide in Houttuynia cordata Thunb.

Abstract

The extraction in polysaccharide in Houttuynia cordata was studied.The results showed that the best extraction conditions were:heating circumfluence for 2 h and for 3 times,the ratio of water to solid was 30∶1.The samples were precipitated by ethanol to remove protein and then separated by gel chromatography.The samples were extracted with water by ultrasonic oscillation and by solid phase extraction with Sep-pak C18 cartridge.Sugars were separatedusing Waters high performance carbohydrate column as solid phase,7∶3 with acetonitrile∶water as mobile phase,and detected with evaporative light-scatter detector.The component of polysaccharides of Houttuynia cordata was m(wood sugar)∶m(fructose)∶m(arabinose)∶m(galactose)=2.209∶1.587∶1.000∶2.092.
