Abstract
Aim:To investigate the effects of glycine on the expression of TNFα and IL 10 in peritoneal macrophages of rats. Methods: The isolated peritoneal macrophages from Wistar rats were cultured in RPMI 1640 medium supplemented with 10% fetal bovine serum or in RPMI 1640 with 2 mmol/L glycine and 10% fetal bovine serum. After LPS(5 mg/L) stimulation, both the TNFα and IL 10 levels in supernatants at 0, 3, 6, 12 and 24 hours and the mRNA expression of TNFα and IL 10 at 12 hours were detected. Results:a) Glycine (2 mmol/L) significantly reduced TNFα level at 3 hours ( P 0.05), 6 hours ( P 0.01) and 12 hours ( P 0.01).In addition, glycine inhibited 12 hour TNFα mRNA transcription after LPS sitmulation ( P 0.01).b) Glycine(2 mmol/L) significantly increased LPS induced IL 10 production at 6 hours ( P 0.05) and 12 hours ( P 0.001). Glycine accelerated 12 hour IL 10 mRNA expression after LPS stimulation ( P 0.05).Conclusion: These data indicate that glycine time dependently inhibits LPS induced TNFα production and increases IL 10 production at both transcription and translation levels.
