EFFECTS OF NORADRENALIN ON TNF-α AND IL-1β EXPRESSIONS IN RAT KUPFFER CELLS INDUCED BY LPS

Abstract

Objective To evaluate the effect of noradrenalin on the expression of TNF-αand IL-1βin rat Kupffer cells induced by lipopolysaccharide(LPS).Methods The isolated rat kupffer cells cultured in vitro divided into three groups,Given lipopolysaccharide(10 μg/ml)with or without norepinephrine(0.1 μmol/L~10 μmol/L)respectively for 12 hours.Using quantitative reverse transcription-polymerase chain reaction to detect TNF-α and IL-1β mRNA expression,ELISA detect TNF-α,IL-1β protein expression of cell culture supernatant.Results In the LPS and norepinephrine(1 μmol/L and 10 μmol/L)group,the cell culture supernatant TNF-α and IL-1β protein were significantly higher than that in LPS group(P0.05);TNF——α mRNA expression were higher than that in LPS group 50.9%(P0.05)and 59.1%(P0.05)Respectively;IL-1β mRNA expression increased 53.7% and 57.8% than that in LPS group(P0.05).Conclusion The high concentration of noradrenalin could enhance the expressions of TNF-α and IL-1β of hepatic Kupffer cells induced by lipopolysaccharide and has pro-inflammatory effects.
