Abstract

Objective To explore the effects of nuclear-factor-κB inhibitor,PDTC,on the content changes of the tumor necrosis factor-α(TNF-α) in plasma and BALF in rats with acute lung injury(ALI) induced by lipopolysaccharide(LPS).Methods 54 Male rats were randomly divided into 3 groups. The control group was made up of 6 rats(Group N,n =6) which were injected with 8 mL/kg normal saline into the peritoneal cavity.Group ALI consisting of 24 rats(Group L,n = 24) were injected with 8 mg / kg LPS,and in Group PDTC(P + L group,n = 24) the rats were injected with 120 mg / kg PDTC and 8 mg / kg LPS,with an interval of half an hour. The samples of group L and group P + L were observed to track the changes of TNF-α in blood plasma and the bronchoalveolar lavage fluid(BALF) at 2 h,4 h,8 h and 12 h.Results Compared with that in the control group,TNF-α increased significantly in Group L(P 0.01) but decreased significantly compared with that of Group P + L(P 0.05).Conclusion LPS has caused a great amount of TNF-α release in blood plasma and BALF.It suggests that NF-κB is involved in control of inflammation and plays an important role in ALI.PDTC can control the expression and release of inflammation, and thereby effectively relieve the ALI induced by LPS in rats.
