Abstract
Objective To explore the effects of nuclear-factor-κB inhibitor,PDTC,on the content changes of the tumor necrosis factor-α(TNF-α) in plasma and BALF in rats with acute lung injury(ALI) induced by lipopolysaccharide(LPS).Methods 54 Male rats were randomly divided into 3 groups.The control group was made up of 6 rats(Group N,n =6) which were injected with 8 mL/kg normal saline into the peritoneal cavity.Group ALI consisting of 24 rats(Group L,n = 24) were injected with 8 mg / kg LPS,and in Group PDTC(P + L group,n = 24) the rats were injected with 120 mg / kg PDTC and 8 mg / kg LPS,with an interval of half an hour.The samples of group L and group P + L were observed to track the changes of TNF-α in blood plasma and the bronchoalveolar lavage fluid(BALF) at 2 h,4 h,8 h and 12 h.Results Compared with that in the control group,TNF-α increased significantly in Group L(P 0.01) but decreased significantly compared with that of Group P + L(P 0.05).Conclusion LPS has caused a great amount of TNF-α release in blood plasma and BALF.It suggests that NF-κB is involved in control of inflammation and plays an important role in ALI.PDTC can control the expression and release of inflammation,and thereby effectively relieve the ALI induced by LPS in rats.
