Abstract

【Objective】 To explore the effects of Resveratrol on TRAIL-induced apoptosis of MDA-MB-231 breast cancer cell line.【Methods】 The proliferation was detected by MTT assay.Apoptosis of cells was analyzed by Annexin V staining combined with flow cytometry assay.The relative activity of Caspase-8 and Caspase-3 were examined by spectrophotometric method.Nuclear was stained with DAPI and its morphology was observed.【Results】 Exposure of cell to 50 ng/mL TRAIL combined with 50 μmol/L and 100 μmol/L Resveratrol for 12 h increased inhibitory effect on cell proliferation which were 62.43±2.07%,87.32±1.44% respectively,as well as enhanced the apoptotic percentage which were 53.59±1.44% and 73.72±1.96% separately,compared with that of the cells incubated with TRAIL or Resveratrol alone.The activity of Caspase-8 and Caspase-3 in the cells treated with TRAIL combined with Resveratrol was also much higher than that of cells treated with TRAIL or Resveratrol alone.【Conclusion】 Resveratrol could enhance the effect of TRAIL on proliferation inhibitory and apoptosis promotion of breast cancer cells via increase the expression of Caspase-8 and Caspase-3.
