Abstract
In order to explore the possible mechanism of apoptosis effect induced by resveratrol in human acute leukemia KG-1 cells,the suppressive effect of resveratrol on proliferation of KG-1 cells was analyzed with MTT method.Wring-Giemsa Staining,transmission electron microscope technique were used to detect the apoptosis status of KG-1 cells.The cell cycle and apoptosis percentage of KG-1 cells treated with resveratrol were detected by flow cytometry.The expression of bcl-2 and caspase-3 mRNA was assessed by semi-quantitative RT-PCR.The results showed that resveratrol can obviously inhibit proliferation of KG-1 cells(P0.01).Compared to control group,the cell number in S phase of KG-1 cells treated with resveratrol was increased(P0.01),and the apoptosis rate increased significantly(P0.01) and the expression of bcl-2 reduced,caspase-3 increased(P0.05).Resveratrol has significant inhibitory effect in KG-1 cells in vitro,it is probably related to restrain the expression of bcl-2 and improve the expression of caspase-3.
