Abstract
Objective: To investigate the proliferation inhibition effect of FTY720 on human pancreatic cancer Miopac2 cell strain in vitro. Methods: MTT assay was used to measured the proliferative inhibition effect of FTY720.Flow cytometry was used to detect the effect of FTY720 on cell cycle and apoptotic rate. Morphologic changes were observed by invert microscope. Results: The light absorption value of Miopac2 cells decreased with increasing of the concentration of FTY720 in experinaental groups in MTT test. The re- sult of flow cytometry demonstrated that FTY720 induced Miopac2 cells arrest in G1 phase and apoptosis with dose dependent. The apoptotic cells were found by microscopy. Conclusion: FTY720 inhibits the growth of Miopac2 cells with concentration dependent, which may be correlated to cell cycle arrest and cell apoptosis. It suggests that FTY720 has an antitumor effect.
