Abstract

[Objective]The aim of the research was to discuss the technical links in the tissue culture and rapid propagation of Ruichang yam.[Method]With the stem segments of Ruichang yam as explants,the effects of different hormones and their concn.on callus formation,tufted-bud induction and rooting were researched through tissue culture experiments.[Result]The disinfection efficacy of combing 70% alcohol with 0.1% HgCl_2 on the explants was best.The axillary buds grew best in the medium MS+6-BA 2.0 mg/L+NAA 0.2 mg/L,the induction rates of axillary buds in the media MS+6-BA 1.0 mg/L+NAA 0.2 mg/L,MS+6-BA 1.0 mg/L+NAA 0.5 mg/L,MS+6-BA 2.0 mg/L+NAA 0.2 mg/L and MS+6-BA 2.0 mg/L+NAA 0.5 mg/L were 52%,43%,84% and 48% resp.In subculture,the efficacy of inducing tufted-bud with the original induction medium was worse and reducing the concn.of 6-BA was helpful to tufted-bud induction.The rooting efficacy of seedlings was better in the medium 1/2 MS+IBA 1.5 mg/L+NAA 0.3 mg/L+active carbon 0.2 mg/L.[Conclusion]The research laid a basis for the application of tissue culture technology in seedling breeding.
