Abstract
Objective To study the synergistic effects of polyamine analogue(bis-ethylnorspermine,BENS)combined with aspirin on the apoptosis of cervical cancer Caski cell lines.Methods Human cervical cancer Caski cell lines cultivated in vitro were administered with BENS combined with aspirin,and then MTT method was used to assay the proliferation of Caski cells.Sub-G1 / flow cytometry and DNA fragmentation assays were used to determine the apoptosis.The expressions of apoptosis-related proteins,such as Bcl-2,Bax and cyt-C were measured by Western blot,and the enzymatic activity of spermine oxidase(SMO)was determined by chemical analysis.Results The combination of BENS and aspirin could increase the apoptosis rate of Caski cells markedly(P 0.05),and low dose of aspirin could markedly increased the apoptosis-inducing ability of BENS on Caski cells(P 0.01).Under the combination of BENS and aspirin,the ratio of cells in G0 / G1 phase was increased,whilst those in S and G2 / M phase were decreased;the expression of Bcl-2 was decreased,that of Bax was increased,and cyt-C was released from mitochondria.The activity of SMO was not changed significantly.Conclusion Aspirin could markedly increase the apoptosis-inducing ability of BENS on Caski cells,which was not related to the activity of SMO.
