Abstract

Objective:To establish a ultra performance liquid chromatography(UPLC) method for simultaneous determination of 5 lipophilic components in Salvia miltiorrhiza.Methods:The analyses were performed on a Waters Acquity UPLC system,equipped with a binary solvent manager,a sampler manager,a column compartment,and a PDA detector,connected to Waters Empower 2 software.An Acquity UPLC BEH C18 column (50 mm×2.1 mm,1.7 μm) was used for all analyses.The investigated compounds were separated with gradient mobile phase consisting of water and acetonitrile.The temperature of sample manager was set at 35 ℃,and the injection volume was 2 μL.Peaks were detected at 280 nm.Result:The investigated compounds including dihydrotanshinoneⅠ,przewalskin,crytotanshinone,tanshinoneⅠ,tanshinoneⅡA had good linearity(r≥0.9997) over the tested ranges.The average recovery was 98.5% to 103.1% with RSD≤4.2%.Conclusion:The developed UPLC method is simple,sensitive and accurate with good repeatability,which is available for quality control of Salvia miltiorrhiza.
