Abstract

Objective To investigate the correlation between Pro12Ala polymorphism in peroxisome proliferator-activated receptor-γ2(PPARγ2) gene and type 2 diabetes in Guangdong population. Methods Polymerase chain reaction(PCR)-direct sequencing assay was used to determine the distribution of allele and genotype frequencies of Pro12Ala polymorphism in PPARγ2 gene in 143 type 2 diabetic patients and 150 normal control subjects. Results The PP,PA and AA genotype frequencies of Pro12Ala polymorphism in 2 type diabetes and control group were 93.0%,7.0%,0% and 90.7%,9.3%,0% respectively.Frequencies of P allele and A allele were 96.5% vs 95.3% and 3.5% vs 4.7% in type 2 diabetes and control groups respectively. There were no differences in genotype and allele frequencies between the two groups(P0.05).Although the level of fasting blood sugar(FBG),total cholesterol(TC),triglyceride(TG) and body mass index(BMI) in 2 type diabetes with PA genotype was higher than those PP genotype patients,no significant difference was found.(P0.05).Conclusion Pro12Ala polymorphism in PPARγ2 gene is not significantly associated with type 2 diabetes in Guangdong population.
