Effect of Rupatadine on Acute Lung Injury Induced by Oleic Acid in Rats

Abstract

OBJECTIVE To investigate the effect of rupatadine on acute lung injury induced by oleic acid in rats.METHODS Rats were randomly divided into six groups.The changes of lung inspiratory resistance,expiratory resistance and dynamic lung compliance were recorded.BALF(bronchoalveolar lavage fluid)protein was measured by spectrophotometer and TNF-αlevel was examined by En- zyme linked immunosorbent assay(ELISA).Lung wet tu dry weight ratio was calculated to assess alveolar edema.RESULTS The pre- treatment of rupatadine significantly alleviated oleic acid-induced lung injury accompanied by the imprnvement of lung function aud reduc- tion of lung wet tn dry weight ratio.Furthermore,rupatadine also decrcased the concentration of protein in BALF and inhibited the in- crease of the TNF-αsignificantly.CONCLUSION Rupatadine has a dose-depeudent beneficial effect on acute lung injury induced by oleic acid in rats.The ultimate reduction of inflammatory factor was involved,at least in part,in the mechanism of rupatadine effects.
