Protective effect of tyrphostinA1 on oleic acid-induced acute lung injury in rats

Abstract

AIM: To investigate the effect of tyrphostinA1 on acute lung injury induced by oleic acid in Sprague-Dawley rats.METHODS: Acute lung injury was induced by oleic acid in Sprague-Dawley Rats.The changes of lung inspiratory resistance and dynamic lung compliance were real time recorded by anrise2005 pulmonary function test apparatus.Light microscopy and electron microscopy were used to examine histological changes.The lung index and wet to dry weight ratio were calculated.Pulmonary vascular permeability and the concentration of protein in the bronchoalveolar lavage fluid(BALF) was measured by spectrophotometer.The levels of IL-10,IL-6 and TNF-α were examined by enzyme linked immunosorbent assay(ELISA).RESULTS: TyrphostinA1 significantly alleviated oleic acid induced lung injury followed by reduction of lung index and wet to dry weight ratio,improved pulmonary function,decreased pulmonary permeability and protein,TNF-α,IL-6 level in bronchoalveolar lavage fluid,also elevated IL-10 level in bronchoalveolar lavage fluid.Light microscope findings indicated that the impairment in tyrphostinA1 groups was far less severe than that in model groups.CONCLUSION: TyrphostinA1 shows preferable effect on acute lung injury induced by oleic acid in rats.
