Prevention of oleic acid-induced acute lung injury of rats with oxymatrine

Abstract

Objective:To assess the prophylaxis of oleic acid-induced acute lung injury (OA- ALI) in rats with oxymatrine (OMT).Methods:A model of rat acute lung injury was set up by adminis- tering oleic acid into rats.A real-time dynamic record of lung volume and lung compliance with the in- duction of the lung injury was conducted by the AniRes2005 Pulmonary Function Test Apparatus.The lung histopathological examination was performed by light microscopy.The ratio of lung wet/dry weight was determined when the lung tissues were collected from the tested rats under euthanasia.Serum TNF-αlevel of the rats was measured by enzyme-linked immunosorbent assay (ELISA).Results:OMT im- proved lung function of rats with OA-ALI,manifesting less histopathological changes,lower ratio of lung wet/dry weight and reduced serum TNF-αlevel.The inhibition rate of OMT at 30,60 and 90 mg·kg~(-1) was 19.9%,30.9% (P<0.05) and 43.9% (P<0.01),respectively.Conclusion:OMT has a dose- dependent prophylaxis of rats from OA-induced acute lung injury by inhibition of serum TNF-αlevel.
