Abstract
Insulin resistance is one of the main mechanisms of type Ⅱ diabetes,it leads to hyperglycemia,and ultimately leads to diabetes.The current insulin-resistance cell models are mainly single factor-induced models.Induced HepG2 cell model using elevated glucose and free fatty acids seems more in accordance with the high cholesterol and hyperglycemia status of human body when the disease occurs.To determine the reasonable modeling condition,cells were induced by 25 mmol/L glucose combined with different concentrations of oleic acid and palmitic acid(molar ratio 2:1) for 36 h,later the glucose absorption,glycogen and triglyceride levels were tested.HepG2 cell model of insulin resistance was established with 0.5 mmol/L free fatty acids,and four components were evaluated.The results showed that puerarin,stevioside,ursolic acid and epicatechin could effectively alleviate the insulin resistance.Moreover,stevioside and epicatechin had better effects.
