Abstract

Objective To observe the influence of free fatty acids(FFA)on glucose uptake and insulin sensitivity of 3T3-L1 adipocytes and establish the FFA-induced insulin resistance cell model.Methods 3T3-L1 cells were induced into adipocytes identified by oil red O staining,specific glucose transport was determined in different concentrations of palmitic acid(PA)and oleic acid(OA)induction.Results 3T3-L1 adipocytes induction rate averaged 98%±1.3% by Student method.Specific glucose transport by PA and OA appeared a time and concentration-dependent downward trend after insulin stimulation,significantly lower than the control(p0.01).Specific glucose transport of the basic state decreased significantly 24h after 0.5 mM PA induction compared with the control and 0.25 mM PA induction for 24h(p0.05). Conclusion 3T3-L1 cell can be induced into mature fat cells in vitro,the FFA-induced 3T3-L1 adipocytes produce insulin resistance cell model.Insulin-stimulated specific glucose transport was inhibited in a concentration and time- dependent manner,while that of the basic state was inhibited at high concentration FFA induction.
