The inhibition of IL-6 and IL-8 release from human umbilical vein endothelial cells by endostatin

Abstract

Objectives:To investigate the inhibitory effects of endostatin on IL 6 and IL 8 release from human umbilical vein endostelial cells(HUVEC). Methods: HUVEC culture model was founded. The human endothelial culture supernatant(HECS) was harvested from primary culture medium for 1~9 days and was centrifuged. The cells were divided into two groups: normal control and endostatin. IL 6 and IL 8 in HECS was measured using sandwich enzyme linked immunosorbent assay(ELISA)method. Results: IL 6 and IL 8 were detected in HECS without stimulating factors in the endostelial culture. The amounts of IL 6 and IL 8 increased along with the cell culture time. At 6th day the amount reached their peaks (2 979.32±19.65)pg/ml and (6 018.87± 56.74)pg/ml. In the endostatin group, the amounts of IL 6 and IL 8 were decreased. Conclusions: Endostatin could inhibit the growth, reproduction and the biological activity of endothelial cells.
