Establishment and Observation of Type I Diabetic Rat Models

Abstract

Objective To establish Type Ⅰ diabetic rat models by intraperitoneal injection of streptozotocin and observe the stability of diabetic changes in rats.Methods 70 male Wistar rats were divided into 3 experiment groups of 20 rats each and 1 control group of 10 rats.The experiment groups were intraperitoneally injected by 30 mg/kg,65 mg/kg,and 100 mg/kg STZ respectively,while the control group was injected citrate buffer.Bayer blood glucose meter was adopted to test the blood glucose,setting 16.65mmol/L after 7 d as the type 1 diabetic animal model.The fasting blood glucose level,body weight,water intake,insulin and C-peptide of the rats at different time points were observed/measured.Results In the group of injection by 65 mg/kg STZ,after 7 days most rats reached the model blood glucose standard and had diabetic manifestation,after continuous observation of 7 weeks,18 rats were qualified as the model with obvious symptoms and without conversion.Conclusion With the dose of 65 mg/kg of STZ through one intraperitoneal injection,type 1 diabetic rat models can successfully established.
