Abstract

OBJECTIVE To explore the construction of type 2 diabetic animal model.METHODS Thirty five male rats were randomly divided into control group(ten rats)and experimental group(twenty five rats),experimental group were fed with high-carbohydrates-fat diet and the control group were fed with basic diet.One month later,the rats in diabetic group received one-off intraperitoneal injection of streptozotocin at a dose of 25mg/kg.bw,and the control group received intraperitoneal injection of same dose of buffer solution.72 hours later,blood glucose was detected and the rats with blood glucose more than 16.7mmol/L were identified diabetic rats.RESULTS 72 hours later,compared with the control group the blood glucose,serum TG,TC,LDL-C and HDL-C of diabetic group increased significantly(P﹤0.05),and the rats in diabetic group had abnormal glucose tolerance and insulin tolerance,and more drinking,more eating,more urine of type 2 diabetic symptoms.CONCLUSION The type 2 diabetic animal model in rats can be induced by high-carbohydrates-fat diet assisted with low-dose intraperitoneal injection of streptozotocin successfully.
