Effect of Total Phenolics Acid of Oenanthe Javanica on Hepatic Fibrosis in Rats

Abstract

Objective To investigate the protective effect and the possible mechanism of Total Phenolics Acid of Oenanthe Javanica in resisting chronic hepatic fibrosis induced by carbon tetrachloride in rats.Methods A hepatic fibrosis model was induced via hypodermic injection of 40% carbon tetrachloride olive oil solution for 42(0.5ml/100g for the first time,then 0.3ml/100g later on,once every three days).Drugs were administered from the first day an hour before carbon tetrachloride was injected for forty-two consecutive.24 hours after the last administration,the level of hepatic function indices(ALT,AST,ALB,ALP,TP,ChE,LDH)and hepatic fibrosis indices(HA、LN,Ⅲ-C,Ⅳ-C)in serum and MDA content,SOD activity in hepatic tissue were detected.Liver histopathological changes andα-SMA,TGF-β1,TIMP-1 expression quantity in hepatic tissue were also observed.Results The rats in the model group suffered obvious hepatic damage and chronic fibrosis.Liver function and hepatic fibrosis were improved in drug groups,The liver pathological observation showed that total phenolics acid of Oenanthe Javanica could relieve the degree of hepatic fibrosis obviously.Conclusion total phenolics acid of Oenanthe Javanica played an important role in preventing chronic fibrosis of the rat liver.The mechanism may have something to do with the elimination of free radicals and antioxidation and the inhibition of the activation of hepatic stellate cells.
