Establishment and evaluation of pressure overload-induced hypertrophy model in rats

Abstract

Objective To establish and evaluate pressure overload-induced hypertrophy model in rats. Methods Male Sprague-Dawly(SD) rats with body weight of 240~260 g were divided into two groups: left ventricular hypertrophy group(LVH,n=15) and sham group(SHAM,n=15).Pressure overload hypertrophy was induced by aortic abdominal constriction(AAC) by using a silver clip with 0.6 mm internal diameter.Hemodynamics parameters,heart mass index(HMI) and left ventricular mass index(LVMI) were measured after 24 weeks.Pathological changes of left ventricular tissue were observed.Results Compared with SHAM group,systolic arterial pressure(SAP),diastolic arterial pressure(DAP),mean arterial pressure (MAP),left ventricular systolic pressure(LVSP),left ventricular end diastolic pressure(LVEDP) in LVH group were obviously enhanced(P0.01,P0.001) in vivo.HMI and LVMI increased remarkably in LVH group(P0.001).Severe hypertrophic changes were shown in heart tissue of LVH rats by pathology. Conclusion Intermittent pressure overload 24 weeks can trigger abnormity of heart function and morphology in rats.The rat model of pressure overload-induced hypertrophy was established successfully by this way.
