Effect of Shexiang baoxinwan on left ventricular hypertrophy in rats subjected to pressure overload

Abstract

Objective:To investigate the effect of Shexiang Baoxinwan(SXBXW) on left ventricular hypertrophy in rats subjected to pressure overload.Method:Left ventricular hypertrophy was induced by chronic abdominal aortic banding in adult Sprague-Dawley(SD) rats.The rats were classified into SXBXW,metoprolol group(Met),left ventricular hypertrophy group(LVH),sham-operated group(Sham).Rats in SXBXW group and Met group were respectively administered with 13.5 mg/kg of SXBXW and 10 mg/kg of metoprolol for 8 weeks,while rats in LVH group and Sham group received an equivalent volume of 0.9% saline alone for 8 weeks.Echocardiography were done after 8 weeks of treatment,body weight(BW),heart weight(HW),left ventricular weight(LVW),heart weight index(HWI) and left ventricular weight index(LVWI) were recorded.SerumB-type natriuretic peptide(BNP) and alpha tumor necrosis factor(TNF-α) were detected by ELISA.BNP mRNA of the myocardium tissue was detected by RT-PCR.Result:SXBXW's BW has no significant difference among other three groups(P0.05),however its HW was significantly different from LVH group(P0.05) with no difference from Met and Sham groups(P0.05).Compaired with LVH group,LVW,HWI,LVWI and IVSd(diastolic interventricular septal thickness),IVSs(systolic interventricular septal thickness),LVPWd(diastolic left ventricular posterior wall thickness) and LVPWs(systolic left ventricular posterior wall thickness) in group of SXBXW were improved obviously(P0.05),but were still worse than Sham group(P0.05) with no differecen from Met group(P0.05).SXBXW's serum BNP level and myocardium tissue's BNP mRNA level were also much lower than in LVH group(P0.05) while there was no difference among groups of SXBXW,Met and Sham(P0.05).There was no significant difference among each other group in serum TNF-α level(P0.05).Conclusion:SXBXW can reverse pressure-overloaded induced left ventricular hypertrophy,improve ventricular reconstruction,degrade BNP,but has little impact on TNF-α expression,and has no significant difference compaired with metoprolol group.
