Simultaneous HPLC determination of chlorogenic acid and baicalin in Yinhuang preparations

Abstract

Objective:To develop an RP-HPLC method for simultaneous determination of chlorogenic acid and baicalin in Yinhuang preparations.Methods:The chromatographic analysis were carried out on a Waters Symmetry Clscolumn(3.9 mm×150 mm,5μm)with methanol-0.4% phosphoric acid as mobile phase at 0.8 mL·min~(-1) flow rate in gradient mode (0→15 min:methanol 15 %→55%,15→25 min:methanol 55 %,25→28 min:methanol 55%→15%).Results:The linear ranges of chlorogenic acid and baicalin were from 6.88 to 110.08μg·mL~(-1)(r =0.9996) and from 13.40 to 214.40μg·mL~(-1)(r=0.9998),respectively.The mean recoveries of chlorogenic acid and baicalin were 97.4% with RSD 2.2% (n=5) and 96.7% with RSD 2.0% (n=5).Conclusion:The method is simple,fast and accurate.It can be used to control the quality of Chinese herb Yinhuang preparations.
