Clincal Observation in Temozolomide Combined with Radiotherapy in Treatment of Postoperative Malignant Glioma

Objective:To compare the efficiency and safety of temozolomide administered concomitantly and adjuvantly to three dimensional conformal radiotherapy versus three dimensional conformal radiotherapy alone in patients with postoperative malignant glioma.Methods:31 postoperative patients with pathologically confirmed malignant glioma were randomly assigned to receive Temozolomide concomitantly with three dimensional conformal radiotherapy(experiment group,n=13,followed by six cycles of temozolomide);and three dimensional conformal radiotherapy alone(control group,n=18).Results:The overall response rates(CR+PR)were 69.24% in experiment group,included 3(23.08%) patients with complete responses and 6(46.16%)partial responses;and 38.9% in the control group,included 3(16.67%)patients with complete responses and 4(22.23%) partial responses,and with singnificantly difference between the 2 groups,P0.05.The 1-,2-,and 3-year survival rates of patients were 92.13%,79.35%,60.21% in the experiment groupand,58.77%,45.23%,28.63% in the control group.The median recurrent period were 18 months in the experiment group and 15 months in the control group,and with singnificantly difference,and P were 0.033,0.019,0.045 and 0.04 respectively.Conclusion:Three dimensional conformal radiotherapy with temozolomide are more effective than radiotherapy alone in malignant glioma,and have not adverse effect.
