Abstract

Objective To investigate the protective effect of teprenone on gastric mucosal injury due to duodenogastric reflux(DGR)in rats. Methods SD rats were divided into blank group,DGR group, and teprenone-intervention group.A rat model of DGR was established by operation. The DGR group received gastric administration only with 0.9% brine and the teprenone-intervention group received teprenone at a dose of 200 mg/kg daily for 8 weeks. The pathological analysis on the gastric specimens from the rats was then performed to determine the ulcer index(UI)of gastric mucosa and the thickness of gastric gelatinous mucus. The contents of hexosamine and phospholipids in the gastric mucosa were detected and the level of PGE2 was measured by radioimmunoassay. Results The pathological change and UI of gastric mucosa in teprenone-intervention group were significantly lower than those in DGR group(P 0.05),while the thickness of gastric gelatinous mucus, the contents of hexosamine and phospholipids, and the level of PGE2 were markedly higher(P 0.05 or P 0.01).The UI in teprenone-intervention group, however, was higher than that in the blank group and the other indexes were lower(P 0.01).Conclusion Teprenone plays a role in protection against gastric mucosal injury secondary to DGR.
