Abstract

OBJECTIVE:To establish a method for determining the contents of amlodipine besylate and valsartan in compound reserpine.METHODS:The chromatographic column of Agilent Eclipse Plus C18(5 μm,4.6 mm×250 mm) was used with injection volume of 20 μL.The column temperature was maintained at room temperature. The mobile phase consisted of methanol-0.2% phosphoric acid(70∶30) at a flow rate of 1.0 mL·min-1.The detection wavelength was set at 237 nm.RESULTS:The chromatographic peak area of the reference standard and sample volume showed a good linear relationship in a range of 0.017 34-0.173 4 μg for amlodipine besylate and 0.401 5-4.015 μg for valsartan, with correlation coefficient(r) being 0.999 94 and 0.999 99,respectively and average recoveries being 99.2%(RSD=1.29%,n=6) and 99.6%(RSD=0.71%,n=6),respectively.CONCLUSION:This HPLC determination method is fast, simple and reproducible and it can be used in the determination of amlodipine besylate and valsartan in compound reserpine.
