Abstract

Objective To investigate effect of matrine on the proliferation and apoptosis of human ovary cancer A21-2 cells matrine and its molecular mechanisms.MethodsA21-2 cells were treated with matrine at different concentration.Morphology changes of cell apoptosis were observed by transmission electron microscopy. The inhibition of proliferation was determined by tetrazolium bromide colorimetry(MTT) method. The expression of caspase-3 mRNA was analysed by reverse transcription polymerase chain reaction.ResultsThe growth of A21-2 cells were inhibited of in a time-and dose-dependent fashion by matrine. After matrine treatment,A21-2 cells showed apoptosis morphological changes under transmission electron microscopy. And the expression of caspase-3 mRNA was up-regulated(P0.01).ConclusionMatrine can inhibit proliferation and induce apoptosis of A21-2 cells. It's molecular mechanism might be related to modulating the expression of caspase-3.
