Preparation of polyclonal antiserum of rabbit anti-rat sodium channel Nav1.8

Abstract

Objective To obtain rabbit anti-rat polyclonal antiserum. Methods The short peptides from C terminal of rat Nav1.8 were synthesized and New Zealand white rabbits were immunized by this antigen to obtain polyclone antiserum. The antibody was purified ammonium sulfate.The titer of the antibody was assayed by the experiment of agarose biphasic diffusion. Results Antiserum against Nav1.8 was obtained in immunized rabbits. Immunohistochemical test with this antibody showed that this antibody was expressed in the affected nerves of the patients with TN. Conclusion The rabbit anti-rat polyclonal antiserum can replace the antibody of hNav1.8 to be applied.
