Expressions of PTEN,PI3K and Akt Protein in Gastric Carcinoma and Their Clinical Significance

Abstract

Objective: To investigate the expressions of PTEN,PI3K and Akt proteins in gastric carcinoma and their relationship between their expressions and clinical pathology parameters,and correlation among PTEN,PI3K and Akt protein expression each other.Methods: Elivision immunohistochemical method was used to detect expressions of PTEN,PI3K and Akt proteins in ten normal gastric mucosa tissues and 68 gastric carcinoma tissues.Results: In 68 gastric carcinoma tissues,PTEN,PI3K and Akt proteins positive rates was 48.53%(33/68),85.29%(58/68) and 89.71%(61/68) respectively.There were significant differences of PTEN,PI3K and Akt proteins expressions between gastric carcinoma and normal gastric mucosa epithelium(all P(0.05).) There were significant correlation between PTEN,PI3K and Akt proteins expression and lymph node metastasis or cell differentiation degree respectively(all P0.05).Moreover,significant correlation was detected between positive rates of PTEN protein and Lauren type of gastric carcinoma(P=0.022).In 68 cases of gastric carcinoma,a negative correlation was found between the expression of PTEN and PI3K or Akt proteins respectively(both P0.05),and there was a highly positive correlation between PI3K and Akt protein expression(P=0.001).Conclusion: All of PTEN,PI3K and Akt proteins expressions were related with the development,progress,invasiveness and metastasis of gastric carcinoma,and PTEN was related closely with the development of diffuse-type gastric carcinoma.In the development and progress of gastric carcinoma,PI3K and PTEN may counteract each other,and the loss of PTEN protein expression may be related with the overexpression of Akt protein.PI3K and Akt protein may cooperate to promote malignant progress of gastric carcinoma.
