Effects of Privet Tang Maipingfang on mRNA Expression of Blood Vessel Endothelium Monocyte Chemoattractant Protein-1 in Type 2 Diabetic Rats

Abstract

Objective To investigate the expression of blood vessel endothelium monocyte chemoattractant protein-1 (MCP-1) in type 2 diabetic rats following the treatment of Chinese medicine Tang maipingfang.Methods The type 2 diabetic Wistar rats were established by injecting high dose (60 mg/kg) of streptozotocin (STZ), and the successfully established rat models were randomly divided into model group (n=8), Privet Tang maipingfang group (n=11), Metformin group (n=11) and Yuquanwan group (n=10). Another 10 normal Wistar rats served as normal control. Their body weight and blood glucose were measured every two weeks. At the end of six weeks, all rats were sacrificed and their aortas were taken to detect the expression of MCP-1 by semi-quantitative RT-PCR.Results After successful establishment of rat model of type 2 diabetes, their blood glucose rose and their body weight decreased. After treatment, the expressions of MCP-1 in Privet Tang maipingfang group and Metformin group were significantly lower than that of model group (P0.01).Conclusion Privet Tang maipingfang can decrease the expression of MCP-1 mRNA levels, abate inflammatory reaction of blood vessel endothelium, restrain and decrease arteriosclerotic formation.
