Abstract
[Objective] To investigate the effect of medroxyprogesterone acetate (MPA) combined with cisplatin (DDP) on the invasion of drug-resistant human ovarian cancer cell line CoC1/cDDP and the mechanicsms.[Methods] The invasiveability,the survivin-ΔEx3 mRNA and Caspase-3 mRNA were measured by RT-PCR,the G_1 cells and ap- optosis cells were measured by FCM.[Results]The invasive ability,apoptosis rate and the survivin-ΔEx3 mRNA,Caspase -3 mRNA expression of CoC1/cDDP had no significant changes after being treated with 10μg/ml DDP for 48 h(P0. 05).The abilities of MPA+DDP treated and 20μg/ml DDP treated cells to invade the reconstituted basement membrane were decreased significantly.The apoptosis rate was increased.The expression of survivin-ΔEx3 mRNA was decreased(P0.05),but the expression of Caspase-3 mRNA was increased(P0.05),the changes were in a dose-dependent manner(P0.05).[Conclusions] MPA has strong reversal effects on DDP resistance in CoC1/cDDP,has inhibitory effect on the invasion of CoC1/cDDP and induces apoptosis of CoC1/cDDP by down-regulating survivin-ΔEx3 mRNA ex- pression and increasing Caspase-3 mRNA expression.
