Expression of HIF-1α in placent and vascular endothdial growth factor,soluble fms-like tyrosine kinase receptor-1 in serum and urinary: preliminary pathophysiological reasearch of preeclampsia

Abstract

Objective To investigate the expression of hypoxia-inducible factors-1α in placenta and serum vascular endothdial growth factor,soluble fms-like tyrosine kinase receptor-1 in maternal serum and urinary from normal pregnant women and preeclampsia,so as to probe the possible mechanism of preeclampsia. Methods Placentas,serum and urinary were collected from 50 preeclampsia women including 22 mild preeclampsia and 28 severe preeclampsia as study group and 30 normal pregnant women as control group. Expressions of HIF-1α were detected by Western blot. Cellular location was determined by immunohistochemistry. The levels of VEGF and sFlt-1 in serum and urinary were detected by enzyme linked immunosorbent assay. Results( 1) The expressions of HIF-1α in placental villous syncytiontrophoblast of preeclampsia group were significantly higher than that of control group.( 2) The level of VEGF in serum and urinary of the mild preeclampsia group and the sever group were significantly lower than that of control group( P 0. 01). The level of sFlt-1 in serum and urinary of the the mild preeclampsia group and the sever group were significantly higher than that of control group( P 0. 01).( 3) There were positively correlation between he expressions of HIF-1α in placental and the level of sFlt-1 in serum and urinary of study group. Conclusion HIF-1α and its target gene VEGF,sFlt-1 not only paticipate in pathogenic process of preeclampsia but also are related to its degree.
