Abstract
Objective To evaluate the changes in platelet function after filtering leukocytes with two types of leukocytes filter for PCs.Methods PCs was prepared from 400 ml fresh whole blood by platelet-rich plasma(PRP) method.Six bags of ABO-matched PCs were pooled,and then filtered with two types of domestic filters(namely group A and B),10 samples respectively.The counted numbers of platelet and leukocyte,pH,CD62p expression(%),platelet aggregation and hypotonic shock response(HSR) were detected before and after filtering.Results After filtering leukocytes,the platelet recovery,the residual leukocytes and the pH,group A and B respectively,were(87.01±3.47)% vs(87.88±4.77)%,(0.95±0.90)×106 vs(0.45±0.58)×106 and(7.13±0.13) vs(6.80±0.26).The CD62p expression(%),platelet aggregation and hypotonic shock response(HSR) of group A before and after filtering were(8.06±4.11)% vs(8.21±4.50)%,(70.55±27.21)% vs(71.63±32.24)% and(68.14±10.13)% vs(69.18±9.38)%,while those of group B were(10.34±3.26)% vs(10.47±2.42)%,(56.30±18.43)% vs(59.49±19.15)% and(75.73±5.50) vs(73.74±6.52)%,respectively.There were no significant difference(P0.05)between these items before and after filtering.Conclusion Filtration with two types of domestic leukocyte filters didn′t activate platelet and had no influence on platelet aggregation and HSR.Besides,both the platelet recovery and residual leukocytes met the requirements of Chinese standards.These two types of domestic leukocyte filters could be applicable to the leukocyte removing for PCs.
