Study of Platelet Function Following Concentration after Leukocytes Filtration

Abstract

Objective To evaluate the effect of platelet concentration after leukocytes filtration. MethodsConcentrated platelets were prepared from 400 ml fresh peripheral blood by platelet-rich plasma(PRP)method.Six bags of ABO-matched platelets were pooled and filtered. The number of platelets and leukocytes were counted,and pH,CD62p expression(%),platelet aggregation and hypotonic shock response(HSR) were detected before and after filtration.Results The filtered leukocytes and platelet recovery was(87.46± 3.56)%,and the residual leukocytes was(98.61± 1.03)%.The CD62p expression(%) before and after filtration was(9.63± 3.86)% and(9.72± 3.91)%(P0.05), respectively.Platelet aggregation was(57.44± 12.58)% and(59.28± 15.23)%(P0.05). Hypotonic shock response was(75.83±8.67)% and(73.42± 7.24)%(P0.05).No significant effect of filtration was noted on the founctions of leukocytes and platlets.Conclusion Filtration with leukocyte filters will not activate platelet and have no influence on platelet aggregation and HSR. Besides,both platelet recovery and residual leukocytes met the requirements of Chinese standards.
