Abstract

Objective To investigate the effects of different doses of angiotensin Ⅱ(Ang Ⅱ) on proliferation and concentration of intracellular free calcium of rat hepatic stellate cells(HSC) and to study the correlation between proliferation of HSC and intracellular free calcium content. Methods HSC were isolated and cultured. Proliferation and intracellular free calcium concentration were measured by the MTT method and flurescence measurement with Fura-2/AM. Results It showed that 10 -9 ~10 -5mol/L Ang Ⅱcould significantly increase intracellular free calcium content of HSC in a dose-dependent style;10 -9 ~10 -6mol/L Ang Ⅱ could significantly promote proliferation of HSC(P0.05),but 10 -5mol/L Ang Ⅱ could not significantly promote proliferation of HSC(P0.05). Conclusion Ang Ⅱ could increase intracellular free calcium content in a dose-dependent style and promote HSC proliferation which being certain in the range of appropriate dose, correlativity.
