Abstract
The hydrolysis of perilla leaves by cellulase for the extraction of polysaccharides was optimized using response surface methodology.Higher extraction efficiency of polysaccharides from vacuum dried perilla leaves was obtained when compared to those dried by other methods.One-factor-at-a-time method was used to investigate the effects of enzyme dosage, temperature and hydrolysis time on the extraction efficiency of polysaccharides, and a second-order polynomial regression model was established using a Box-Behnken experimental design. The optimum extraction conditions were determined to be 1771.85 U/g,53.7C and 36.2 min for enzyme dosage, temperature and hydrolysis time, respectively.Under these conditions, good agreement between the predicted(18.21 mg/g) and measured(17.91 mg/g) extraction efficiencies of polysaccharides from perilla leaves was observed.
