Influence of Sijunzi Decoction on Gastric PCD and Caspase-3 Expression in Spleen Deficient Rats

Abstract

Objective To discuss gastric cell death(PCD)changes in the spleen deficient rat models,and the influence of Chinese medicine Sijunzi decoction on the changes. Methods A total of 102 SD rats were randomly divided into the normal control group(n =24),the long-term model group(n =26),the natural recovery group(n =26)and the Sijunzi decoction group(n =26). Rats in the normal control group were raised in the routine way for 7 weeks:they were given normal saline 1.5 mL/100 g by gavage once every other day during the first 4 weeks,and then everyday in the last 3 weeks. Spleen deficiency rat model was made by composite factors in the other three groups. After 4-week modeling,the rats in the long-term group were modeled for another 3 weeks. Rats in the natural recovery group were raised in the routine way for 3 weeks. Rats in the Sijunzi decoction group were given Sijunzi decoction 1.5 mL/100 g by gavage once everyday for 3 weeks. Tunnel assay was used to observe the programmed cell death level and immunohistochemistry was used to evaluate Caspase-3 expression in rat stomach tissues. Results The Caspase-3 content and epithelial cell death level in Sijunzi decoction group decreased(P 0.05). Compared with those in the long-term model group and natural recovery group,the difference was significant(P 0.05). However,the levels are higher than those of normal control group(P 0.05). Conclusion Sijunzi decoction can reduce the chronic stomach diseases by improving the gastric mucosal barrier function.
