Abstract

From June to July, the new semi-ligneous stem nodes of Toona sinensis with axillary buds was taken as explant cultared in vitro to induce axillary bud sprouting and bud multiplications. Medium supplemented with different plant bormones and concentrations of them were required for different stage of the culture. These are as follows 1. The medium for germination:MS+6-BA 0. 2mg/L. Buds of the explants sprouted early and grew fast in this medium. 2. The medium for subculture and shoot multiplications:MS+6-BA 0.2mg/L+GA3 2.0mg/L (first option) or MS+ZT 0. 2 mg/L+GA, 2.0mg/L(second option). The propagation rates were 3.1~4.0 within 30d. 3. The medium for rooting: MS (without inorganic elements) +IBA 1. 0mg/L. The rooting rates was 73%and the quality of the roots was grown well. The survival rate of the plantlet was 72%after transplanting under suitable condition.
