Effects of Danzhi Jiangtang Capsule on function of vascular endothelial cells in rats with type II diabetes mellitus

Objective:To explore the influence of Danzhi Jiangtang Capsule on vascular endothelial function in rats with type Ⅱ diabetes mellitus.Methods:Ninety male SD rats were randomly divided into six groups as control group,model group,high,middle and low dosage of Danzhi Jiangtang Capsule groups,metformin group and Shenqi Jiangtang granule group.Rats in each group except control group were fed with high fat and high sugar diet and received intraperitoneal injection with low dosage of streptozotocin(STZ) to establish the type 2 diabetes rat models.The concentration of NO,endothelin-1(ET-1) and von willebrand factor(vWF) in plasma of rats were tested by using double antibody sandwich method.The levels of glycosylated hemoglobin were tested by using spectrophotometer method.The morphology of vascular endothelial cells of thoracic aorta of rats was observed by using HE staining.Results:The concentration of NO in plasma of rats was decreased,however,the concentration of ET-1 and vWF was increased,and the tendency was more obvious with the development of disease(P0.01,P0.05).The results of glycosylated hemoglobin and blood glucose monitoring showed that the blood glucose of rats in each treatment group was decreased and controlled effectively(P0.01).The result of HE staining showed that Danzhi Jiangtang Capsule had a repair effect on the damages of endothelial cell.Conclusion:Danzhi Jiangtang Capsule could decrease the blood glucose of rats with type 2.diabetes mellitus and improve vascular endothelia cell function,which had certain prevention and therapeutic effect on rats with type 2.diabetes mellitus.
