Abstract
Objective To explore the differences of the differentially expressed genes in colon tissues of patients with Melanosis coli(MC)and normal controls by cDNA microarray. Methods The mRNA from MC and normal colon tissues were reversely transcribed to cDNA with incorporation of fluorescent dCTP(Cy5 or Cy3)to prepare for hybridization probes.The mixed probes were hybridized with the cDNA microarray.The differential genes in 2 groups were detected by scanning the fluorescent signals. Results All together,93 differentially expressed genes were found in MC and normal colon tissues,of which,53 genes showed increased expression and 40 genes showed decreased expression. Conclusion The result concludes that cDNA microarray technique is a suitable method in screening the differentially expressed genes in tissues,and can be used for understanding pathogenesis in melanosis coli.
