Abstract

Objective:To observe the influence of the apoptosis of cultured MCF-7 cell induced by sodium butyrate.Methods: MCF-7 cells were treated with the final concentrations 0,1.25,2.5,5.0,10.0mmol/L of sodium butyrate for 48h,and analyzed the inhibition ratio of sodium butyrate on cells with MTT.The changes of cell growth were observed under inverted microscope.Flow cytometry,terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL)and agarose gel electrophoresis were used to analyze the apoptosis of cells.Results: Different concentrations of sodium butyrate can inhibit the growth of MCF-7 cells evidently.The cells adherence decreased as increasing of time of treatment under microscope.The cell apoptosis rates were 4.8 %,9.4 %,26.1% and 51.9 % by treatment of different concentrations of sodium butyrate.Some cell bodies were deflated and nucleuses were karyopyknosis and yellow or yellow brown granules.DNA ladders was found on agarose gel electrophoresis from cells DNA under the concentration of 10mmol/l.Conclusion: Sodium butyrate can induce MCF-7 cells apoptosis.
